Its been quite some time since i've last updated. Haha...didn't really go online this whole week. Received a call from ayesha on thusday afternoon. SHE HAS GOT HER BIKE LICENSE!!!! :) congratulations, im so happy for her! She really deserves it, having gone through so much in order to earn this lisence. Yeah yeah, finally, hip hip hurray! Rock on ayesha darling =)

Hmm...this week has been a so so one. Dr Moe came back on tuesday. Haiz...all the stress starts coming in. He asked for a progress report by 4pm the next day. Min Min, hui hua and i expected alot of comments and expectations from him. Turns out we were rite. The first person that he called upon to see her progress was hui hua. He commented so much on her progress, could hear from his tone that he wasn't happy with her. He even asked her to do a powerpoint presentation on her error analysis. Wa...spent around 45 minutes toking to her. Next was min min's turn. For her it went quite ok. But, as usual, he still expected alot from her for the next 2 weeks. Lastly, it was my turn. Previously, he asked to see 20-30 PCR product bands on my next progress report. Ok, i had that. Then i told him i did RFLP on the samples that had bands and showed him my log book. To carry out RFLP, we hav to add 15ul of the PCR product, 2ul of NE Buffer 4, 1ul of NgOMIV and 2 ul of dH2O and run it for 37o celsius for 22 hours. When running the gel for the RFLP product, im suppose to load 20ul of RFLP product mixed with 4ul of 6x loading dye. 10ul of PCR product mixed with 2ul of 6x loading dye before each RFLP product. This was according to wat was written in the previous senior's report. So i jus followed.

And so when i showed him my log book, he said that i am suppose to hav a positive and negative control. So he told me to look through previous people's report, get the sample with the C/C band, and optimise the conditions from there and include this sample on my gel. Since i already used up 35 ul of my samples, i jus do RFLP on the remaining 15ul of PCR product and load into the gel without the PCR product, but including the positive and negative control. But, when i looked through my senior's report and her log book, she didn't do that. So i thought that it was ok not to include that step. But when he looked through my log book and saw that i did not include that step, he told me that i had to include that step. Ok, fine. I was thinking to myself, why do i have to do that when my senior didn;t?

Next morning, which was thursday, i really had no mood to go in. To add on to that, it was raining heavily and i was feeling very cold. And, my menses had to come on that day. Wa...my cramps were really horrible that morning, and because it was so cold in the office, the cramps got worse. I was really mood out that day.

Hmm...anyway, april is ending soon, yeah, now on to May. Hurray! Looking forward to tuesday, movie outing :)

Wow...this week is so relax, dr moe is overseas. Haha...guess wat, there were only two people in the office on thursday, me and the research officer, hui hua. It was super quiet and scary. How i wish everyday was like that :) Went to eat superdog at vivocity with carine and shijin, the 04 girl on thursday. Hmm....i must say, its quite unique. The lemon tea is fragrant, its as if they brewed it themselves. The chicken burger is different, its not the normal chicken burger we eat at other fast food restaurant, but rather, black pepper chicken. And the hotdog is super long. The fries are thicker then the normal one. All in all, i would say that the food there is quite good.


Friday was the last day of attachment for the NTU students. This indian girl was so happy on friday, she couldn't stop laughing. Wish i could be like her:ESCAPE!. Hehe....but too bad i can't, jus started on my project. Aim to finish it by the end of july. Yes, jia you le, u can do it.

I will escape one day. Patience is the key. A little bit of patience and soon i'll be out of there :)

My supervisor is overseas this week. Yeah, so can relax abit. Hehe...gosh submitted my lst progress report today, hmm...know wat was my supervisor's comment? "Not bad but not very good" Coz i have 128 control samples and 97 stroke samples and i onli did 25 samples and out of the 25 samples, only 16 had bands. Haiz...then he wans me to get 20-30 PCR bands on my next progress report in 2 weeks time. Gosh.

Hmm...i wanna sit cable car after my attachment. I wanna go kbox. I wanna shop. I wanna do everything. Haiz....its only the 6th week. And i hav 19 more weeks to go. Starting to get tired and fed up already. And i receive a call from Miss Cherine Tan my liaison officer today. She said she wans me to fix an appointment with dr moe for her to visit me.

Anyone who wans to go shopping on weekends, can call me :)

People are jus so racist, ill-mannered, rude, childish and inconsiderate.

A very common example to show how INCONSIDERATE people can be is in buses and trains. There's so much space inside, but yet people dun wan to move in, all squeeze at the door, making it impossible for other people to board the train. WAT, there are GHOSTS inside izzit?! And then since they dun wan to move in, of course i squeeze myself in, then they give me the black look on the face. HELLO, u were the one who dun wan to move in in the lst place, then when i move in, u give me the black look on the face. WAT, its not my fault ok! Incosiderate n selfish bunch of people.

And then this morning, this RACIST UNCLE pissed me off. My sis and i boarded the bus. There was this 2-seater sit. Since i board the bus lst, i went to bk the 2-seater lst. Walking behind was a chinese uncle, behind the chinese uncle was my sister. Sitting in front of the 2-seater seat was an indian man. He knew that my sister wanted to sit with me, he DELIBERATELY went to sit beside me. My sis and i stared at him, and u know wat, he gave us this SMUG LOOK on the face. Wa...so damn the rude and ill mannered and childish la. I mean, even if he dun wan to sit with the indian man, he doesn't hav to "break up" my sis n i rite? RACIST!

People are jus so rude, ill mannered and inconsiderate.

Anyway, my project has not been going well. Hmm...my PCR is not working well, there are no bands. I think there is too little gDNA in the first place, coz there were only a little left in the vials. Guess i hav to ask them to aliquote for me a new one. Hiaz...and i hav to update my supervisor on monday on my progress, i dunoe wat im gonna tell him man.

Hiaz...pray and hope that all goes well.

Yeah! Started on my project already, finally, after 3 weeks of trying to optimise my PCR conditions. So everyday, i'll basically be doing PCR, gel electrophoresis and restriction fragment length polymorphism (RFLP). The whole process takes 1 day. 1 PCR takes 2 hours, 1 gel electrophoresis takes 1 and a half hours, 1 RFLP takes 22hours. Yup yup, tats how time consuming it is. And there are 128 control samples and 97 stroke samples, so u can imagine how long it will take to finish all the samples.

Im beginning to miss school. Gosh, i never thought that i will miss school. Guess now i understand why it is that working adults say that schooling is better. I feel like going for a ride on the cable cars, i see it everyday when i walked out to eat lunch, gosh, i miss all the fun.

Hmm...been thinking about my future this few days, wondering where i will be in the future. Thinking of going into arts in future. Till then, i shall see how it goes.
